ABSTRACT

ObjectiveTo explore the differences in the protective effects of five formulas for promoting blood circulation and removing blood stasis on the aortic endothelial cells of New Zealand rabbits with heart blood stasis syndrome. MethodEighty New Zealand rabbits were randomly divided into a normal group (n=10) and an experimental group (n=70). The heart blood stasis syndrome model was induced by starvation combined with a high-fat diet and adrenaline in the rabbits of the experimental group. Subsequently, the model rabbits were randomly divided into a model group, a Xuefu Zhuyutang group (3.55 g·kg-1·d-1), a Taohong Siwutang group (2.66 g·kg-1·d-1), a Danshenyin group (1.962 g·kg-1·d-1), a Huoluo Xiaolingdan group (2.80 g·kg-1·d-1), a Shixiaosan group (0.56 g·kg-1·d-1), and a c-Jun N-terminal kinase (JNK) inhibitor (SP600125, 5 μg·kg-1)group. The normal group and the model group received the same amount of distilled water. The rabbits in five Chinese medicine groups were treated correspondingly by gavage, and those in the SP600125 group were injected with 0.5 mL of SP600125-dimethyl sulfoxide diluent. After the treatment, the aorta was collected, and terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay was used to detect the apoptosis of aortic endothelial cells. The enzyme-linked immunosorbent assay (ELISA) was used to detect serum levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6). Western blot was used to detect the protein expression of JNK, phosphorylated JNK (p-JNK), B-cell lymphoma-2 (Bcl-2), Bcl-2-associated X protein (Bax), cysteinyl aspartate-specific protease-9 (Caspase-9), and cysteinyl aspartate-specific protease-3 (Caspase-3) in aortic tissues. Real-time fluorescence-based quantitative polymerase chain reaction (Real-time PCR) was used to detect the mRNA levels of JNK, Bcl-2, Bax, Caspase-9, and Caspase-3 in aortic tissues. ResultFive formulas could improve the apoptosis of aortic endothelial cells to varying degrees. To be specific, Xuefu Zhuyutang and Taohong Siwutang were optimal in efficacy, followed by Huoluo Xiaolingdan, Shixiaosan, and Danshenyin, and SP600125 was the worst (P<0.05, P<0.01). Five formulas could reduce the content of TNF-α and IL-6 (P<0.05, P<0.01), down-regulate the protein expression levels of JNK, p-JNK, Bax, Caspase-9, and Caspase-3 (P<0.05, P<0.01), decrease the mRNA expression levels of JNK, Bax, Caspase-9, and Caspase-3 (P<0.05, P<0.01), and up-regulate the protein and mRNA expression levels of Bcl-2 (P<0.05, P<0.01). ConclusionFive formulas can all reduce the apoptosis of aortic endothelial cells in New Zealand rabbits with heart blood stasis syndrome with different efficacies. It may be related to the different effects of five formulas on the JNK signaling pathway.

ABSTRACT

Since the pathogenesis of depression is complicated, the therapeutic effects of western medicine are poor accompanied by severe side effects. Chinese medicine has unique advantages in the treatment based on syndrome differentiation and contains many effective components against depression, including flavonoids, terpenes, phenylpropanoids, quinones, and alkaloids. These chemical components can delay the course of the disease, improve the curative effect, and reduce side effects of western medicine by regulating the biochemical abnormalities of monoamine neurotransmitters, brain tissue protein content, and internal environment as well as energy metabolism to make the treatment of Chinese medicine highlighted and recognized. This study systematically reviewed the effective components and mechanisms of anti-depressive Chinese medicine to inspire the rational development and utilization of new Chinese medicines against depression.

ABSTRACT

Purpose To investigate the expression and clinical significance of receptor for activated C kinase 1 (RACK1) and β-catenin in cervical cancer tissues. Methods The expression of RACK1 mRNA and β-catenin mRNA in 126 cases of cervical cancer and adjacent normal tissues was detected by RT-PCR, and the expression of RACK1 and β-catenin protein was detected by SP immunohistochemistry. The correlation between the expression of RACK1 and β-catenin protein and clinicopathological data and prognosis was analyzed. Results The relative expression of RACK1 mRNA in cervical cancer tissues was lower than that in adjacent normal tissues, while the relative expression of β-catenin mRNA in cervical cancer tissue was higher than that in adjacent normal tissues. There was a significant difference between the two groups (P < 0.05) . The positive expression rate of RACK1 in cancer tissue group was lower than that in adjacent tissue group (19.0% vs 63.5%) , while the positive expression rate of β-catenin was higher than that in adjacent tissue group (69.0% vs 31.0%) (P < 0.05) . The higher the TNM stage, the lower the differentiation and the lower the RACK1 positive expression rate with lymph node metastasis, the higher the positive expression rate of β-catenin (P < 0.05) .There was a negative correlation between RACK1 and β-catenin protein expression (r = 0.404, P < 0.05) . The survival rates of the patients with strong positive expression of RACK1 protein and weak positive expression of β-catenin protein were higher (42.8% vs 33.3%, 42.8% vs 36.4%) , but the difference was not statistically significant (P> 0.05) . There was a significantly difference in the median survival time between the two groups (57.2 vs 34.0, 44.2 vs 29.8, P < 0.05) . Weak positive expression of RACK1 protein and strong positive expression of β-catenin were risk factors for poor prognosis in patients with cervical cancer (HR were 9.654 and 8.882 respectively, P < 0.05) .Conclusion The abnormal expression of RACK1 and β-catenin may be related to the occurrence, development and prognosis of cervical cancer, and affect the lymph node metastasis of cervical cancer.

ABSTRACT

Object To investigate the changes of neurovascular coupling (NVC) in the contralateral cerebral motor region under the action of repetitive transcranial magnetic stimulation (rTMS) at different frequencies. Methods Some experiments were designed, the electroencephalography (EEG) signals and the near-infrared spectra (NIRS) of the subjects were obtained under different rTMS stimulation frequencies, and EEG power spectrum of each frequency band and NIRS blood oxygen signals to the contralateral movement area were computed and the change characteristics and correlation were analyzed. Results For different stimulation frequencies, the energy of each frequency band of EEG increased after the stimulation, and then decreased rapidly. The oxyhemoglobin (HbO2) concentration was decreased during stimulation, and then raised. The change rate of the two signals varied with different stimulation frequencies, while the change trend was the same. The EEG power spectrum was negatively correlated with the change of HbO2 concentration after stimulation, and it had delayed effect in varying degrees in the period after stimulation. Conclusion The effect of rTMS on the coupling of nerves and vessels in the contralateral cerebral motor region can be reliably revealed by the study of EEG combined with NIRS. This study will be helpful to the development of effective RTMS treatment parameters, and the results are expected to be applied in clinical treatment.

ABSTRACT

Objective To research the influence of the thermoacoustic effect for the frequency of the magnetoacoustic signals in the magnetoacoustic tomography (MAT).Methods Some experiments were designed, and the magnetoacoustic signals and the thermoacoustic signals of several materials with different conductivities were analyzed and compared in the time domain and the frequency domain respectively,of which, copper and graphite were high-conductivity materials, and agar and pork were used as low-conductivity biological or similar tissue. Results The characteristics of the magnetoacoustic signals and the thermoacoustic signals of different materials were acquired. The results illustrated that there was a little drop in frequencies of acoustic signals of the materials with high conductivities while a big drop in frequencies of signals of the tissue-like materials comparing with the stimulant pulse.Furthermore, the main frequencies of acoustic signals were not accordant but much lower than the stimulant ones. Conclusion It is found that the thermoacoustic effect is related to the frequency of the acoustic signals. The drop in the center frequencies of the acoustic signals could be explained by the thermoacoustic effect of materials. The acoustic signals with lower main frequencies have the higher proportion of thermoacoustic signals. The studies could contributing to optimizing MAT method and improving MAT imaging quality.

ABSTRACT

Facing current situation of integrative medicine (IM), authors put forward that clinical and diagnosis program of IM could be carried out from clinical path, pathogenesis, treatment theory and philosophy, and so on, but with different integration degrees. Meanwhile, formulation of concrete program should be disease-targetedly set up, and adjusted from person to person, from place to place, from time to time. As for settled IM program , authors could evaluate it from whether Chinese medicine and Western medicine have formed complementary, synergistic, excitatory actions, and toxicity attenuation; whether more problems could be solved in efficacy, safety, practicability, and economy than previous single mode.
